I'll start us off with one.
This is the first time a member of the main cast takes a holiday in the middle of a story, therefore not appearing in some episodes.

It was the first time (though definitely not the last) they'd spent most of the story more than a few miles away from the TARDIS.
A fairly subtle one. When Barbara finds the damaged Voord submarine, she calls for the Doctor. Up till then, she'd always have called for Ian.
